This is possible, however it is not advised.
Cars that originally come with run flat tyres or RFT's were designed with this in mind. There is no designated space for a spare wheel in the car, and the suspension was designed and fine tuned with the characteristics of a run flat tyre, not a normal tyre.

This can possibly change the feeling of your car in every aspect. such as less responsive turning or altered characteristics in wet weather.
Therefore it is possible, but be aware of the charge in safety as you will not have a spare, and also the change in driving characteristics.
